🚨 New study in @pnas.org ! 🚨

We identified MITD1, a brain-specific interferon-inducible factor that 𝗶𝗻𝗵𝗶𝗯𝗶𝘁𝘀 𝗳𝗹𝗮𝘃𝗶𝘃𝗶𝗿𝘂𝘀 𝗿𝗲𝗽𝗹𝗶𝗰𝗮𝘁𝗶𝗼𝗻, including WNV, USUV, ZIKV & DENV.

Thank you, Juan! I'm glad you enjoyed the read. I believe this small protein still has many secrets to reveal

Thanks for everyone involved in this study, and especially my great superviser who allowed all of this to happen @sebnisole.bsky.social ! 🎊

𝗪𝗵𝗲𝗿𝗲 𝗶𝘀 𝗠𝗜𝗧𝗗𝟭 𝗶𝗻𝗱𝘂𝗰𝗲𝗱 𝗯𝘆 𝗶𝗻𝘁𝗲𝗿𝗳𝗲𝗿𝗼𝗻?

🧠 Mouse data pointed to the 𝗯𝗿𝗮𝗶𝗻, so we tested human brain cells. Only microglia, the immune sentinels of the brain, showed interferon-driven MITD1 expression.

🚨 This makes 𝗠𝗜𝗧𝗗𝟭 𝗮 𝗺𝗶𝗰𝗿𝗼𝗴𝗹𝗶𝗮-𝘀𝗽𝗲𝗰𝗶𝗳𝗶𝗰 𝗜𝗦𝗚, protecting the brain from flaviviruses

𝗛𝗼𝘄 𝗱𝗼𝗲𝘀 𝗠𝗜𝗧𝗗𝟭 𝗯𝗹𝗼𝗰𝗸 𝘃𝗶𝗿𝗮𝗹 𝗿𝗲𝗽𝗹𝗶𝗰𝗮𝘁𝗶𝗼𝗻?

🧬 MITD1 overexpression forms cytoplasmic aggregates that deplete CHMP1B+4B, key ESCRT-III proteins.

🔬 PLA confirmed their recruitment to flavivirus replication factories. Knockdown of CHMP1B+4B hinders MITD1’s antiviral effect—proving a link

To uncover new host factors restricting flavivirus replication, we screened a library of >1,000 interferon-stimulated genes (ISGs) across multiple species.

Among known antiviral ISGs, we found 𝗠𝗜𝗧𝗗𝟭, a never-described-as-antiviral-before factor, as a potent inhibitor of WNV, USUV, ZIKV & DENV

Check our latest opinion piece, just out in @cp-trendsmicrobiol.bsky.social! We delve into how the antiviral RNase ISG20 targets viral RNA and how some viruses escape it.
