I really enjoyed this WIRED article on the prospects of curing pediatric HIV. Grateful to see our work on AAV-vectored delivery of HIV bNAbs featured. I hope it can move toward clinical trials as a potential “one-and-done” strategy to treat HIV infection in children.

I hope this strategy can be evaluated in clinical trials. If this works in humans, it could dramatically simplify efforts to prevent and treat pediatric HIV infections.

In sum, we show that multi-year HIV-1 immunity can be generated in primates from a one-time AAV vector administration at birth–one of the most consistent times of interaction between healthcare providers and the mother-baby pair.

Our study also showed that, compared to adult and juvenile macaques, neonates were more likely to benefit from AAV-vectored delivery of HIV-1 bNAbs because of their tolerance-prone immune systems.

Importantly, neonatal gene therapy with AAV/bNAb vectors significantly protected both infant and pre-adolescent macaques from simian-HIV infection in mucosal challenge models designed to simulate HIV-1 transmission through breastfeeding and sexual intercourse.
