• A 2016 randomized controlled trial (Frye et al., Molecular Psychiatry) found that high-dose leucovorin (2 mg/kg/day up to 50 mg/day) improved verbal communication & adaptive behavior in some children w/ autism, particularly those + for folate receptor antibodies.

Some individuals w/ autism show problems w/ how folate is transported into the brain, particularly due to folate receptor alpha autoantibodies (FRAAs) that block folate transport across the blood-brain barrier. This condition is called cerebral folate deficiency (CFD).
