Thanks so much for the response.. So it has to be the hard way to profile O-glycopeptides.. O-glycoproteases sound interesting.. Didn't know these existed.. Will explore whether these can be of any help.. Cheers..

On a larger note, I guess this is one area where we the mass spectrometrists have to improve communications with the biologists on "the right kind of samples" for mass spectrometry ..

Thanks.. so those scary temperatures and durations are indeed the protocol steps.. one can only hope how good the integrity of proteins after undergoing such treatments..

I was hoping that you will have insights on stored samples.. Thanks for sharing your views.. Shall I take it as don't have high hopes for proteomics with such samples?? 🤔🤔

Thanks for the response.. Good to know that there is precedence.. Also it's nice to have a mental picture of taking brains in buckets to a mass spec lab..

May I ask how the decrosslinking was carried out?? Came across some very scary temperatures and incubation durations suggestions out there..
