Not 100% sure, but I think the issue is that Japanese printers ask for flat tiffs and most foreign publishers get files directly from the printer.

To get layered files, it would mean the artist would have to send two sets, one to the printer and one directly to the publisher abroad.

Not sure about random glyph variation, but you could definitely make a variable font pretty easily if you have something like the Glyphs app.
