The anime's pace is notoriously bad 😭 like there's literally a fan project where they recut and edit it down to be better paced... It's an older series that was weekly and there's some other stuff involved making it like that. Jojo's (anime) is much newer and seasonal.

For example, I have a female friend who talks generally pretty neutral, sometimes a bit fem. But she'll be silly sometimes and say something super masculine for dramatic effect or emphasis. I do something similar too. Most people do, I think.

Using different speech styles from what you'd typically use or what's "normal" can also be seen as humorous. In those cases, it's probably seen as a lot more whatever and normal, because it's "just a gag".

I've had devs ask me to keep gender ambiguous/a mystery/keep players confused, just because. It wasn't even plot relevant lol.
Plus I see JP creators playing around with gender a lot in general.
