FOXA1:p (forkhead box A1) has a single forkhead DNA binding domain just nward of the middle, with a two acceptor phosphoIDR just cward of the middle. Both of the pS acceptors are occupied most of the time. #proteomics

If there is a second DNA-binding domain nward of the homeodomain in the sequence, the active pS acceptors flank this nward domain. Ta-da!

I must admit that taking a look at the phosphorylation patterns associated with transcription factors with homeodomains (homeobox-containing genes) has been kind of a let down. In sequences that rely on a single homeodomain for DNA-binding specificity, the active pS acceptors flank the homeodomain.

I think that from a dose-response point of view, to get the sort of stray hair that you get from a wool sweater you would need to have the pet on the lab bench & be giving it a brush during sample prep.

PKNOX1:p (pre-B-cell leukemia transcription factor/knotted 1 homeobox 1) has the same shiny domain set and arrangement as MEIS proteins, except that its S+phosphoryl acceptors are on the N-flank of the nward DNA interaction domain (MEIS has them on the C-flank). #proteomics

MEIS1:p & MEIS2:p (Myeloid Ecotropic Viral Integration Site homeobox 1 & 2) have 2 bright shiny DNA interaction domains. Both have their S+phosphoryl functional modulation in the pIDR between these domains, with the most prominent acceptors on the C-flank of the nward domain. #proteomics

Everyone in Toronto knows the streetcars are slow and mismanaged (I wish our politicians actually *did* something about this), and yet we spent a generation of transit investments on building what amounts to more of them (which will be poorly run) in places we should have built subways.

Personally, I think a result isn't really worth pursuing unless the underlying protein has been observed in >1000 LC/MS/MS runs & the feature of interest has been observed a few hundred times by multiple groups. But, I am―as everyone knows―more than a bit of an odd ball.
