no worries! this one has been haunting me for a bit. thanks for sharing it!
might have missed it but did you post a solution for this one?
Chicago: if you’re around the southside, the point is a beautiful park around the lake. Also Strings is an amazing ramen place in Chinatown - very easily accessible from the train
Lots more on this project that I cant share quite yet, but special thanks to the Sieber lab at TUM for hosting me in Munich to learn and perform these chemoproteomic experiments! Lipoprotein trafficking is clearly an attractive target for gram(-)’s, especially for beta-lactamase expressing bacteria.
We then demonstrated that OXA-23, the beta-lactamase that confers resistance, is itself an outer-membrane lipoprotein. Inhibiting lipoprotein trafficking via LolF thus also causes over-accumulation of OXA-23 to the inner membrane, which exaggerates these inhibitor’s antibiotic properties.
To understand this unique effect, we used a photoaffinity probe of this HTS hit to assign the Lol complex as the protein target of this compound! Resistance selection, Lol overexpression, and CRISPR studies also confirm lipoprotein trafficking as the major MOA.
Also, an awesome collaborative paper I worked on during my PhD is now published in PNAS!

Our collaborators discovered a small molecule that is a BETTER antibiotic against beta-lactam resistant bacteria than beta-lactam sensitive ones. We wanted to understand why.
ATL Roller Derby Pride match! Highly recommend checking out if there’s a team near you :)

J Med Chem is on 🔥 with the editorials lately, and I highly recommend giving this one a read. Understanding free drug principles is the gateway to all of pharmacology, and is all too often mangled and misapplied by drug discovery practitioners.

literally just had to do this for all my procedures for an SI… add this to the list of things AI could probably do that would be useful 🥲
never used it, but always thought the Beckmann rearrangement was a neat and weird one
Thanks Dr. Rouhier! Hope all is well back on the hill :)
My PhD Defense is tomorrow!! If you’re interested in hearing about some soon-to-be published work on the discovery and synthesis of new gram-negative antibiotic compounds with a unique mechanism of action, DM for a zoom link!
tbh I’m trying to be both at the same time
yikes same, didn’t realize this…
@lmkdassama.bsky.social, who just published an awesome review on gram(-) antibiotic drug discovery!
Corey ylide ftw, love that reagent
Now this is civil disobedience 🔥🔥🔥
Bag of gloves and towels - figured it was something Sn!
