How do SMC complexes load onto DNA to get ready for loop extrusion?

@roisnehamelinf.bsky.social & co discovered that Wadjet, an SMC complex involved in bacterial DNA immunity, performs some impressive molecular gymnastics 🤸‍♂️🤸‍♂️🤸‍♂️.

Thanks for the kind words! It's challenging to acquire sufficient signals for cotranslational folding. I was lucky to find a strong reporter with high expression levels and nice FACS experts to help me. It's quite a journey for sure.

🛡️ I also mapped how molecular chaperones (cellular "helpers") guide folding at specific sites. Trigger factor and DnaK showed varying effects along the full-length protein. This opens new doors for understanding protein misfolding diseases. 🧵4/6

🔍 Surprise finding: Homologous protein domains (EF-G vs EF-Tu) fold through completely different pathways! EF-Tu forms a stable intermediate halfway through synthesis, while EF-G waits until the very end. Structural topology matters! 🧵3/6

⚡ My approach combines SecM arrest peptide, dual fluorescent reporters, cell sorting, and custom deep sequencing to capture folding "snapshots" across entire protein sequences. ⚛ Think of it as a molecular camera that catches the exact moment a protein finds its shape! 🧵2/6

🤔 How do proteins fold while they're still being made? As the main project in my PhD, I developed "Arrest Peptide Profiling" to record proteins folding with as they emerge from ribosomes in living cells 🦠 - with single codon resolution! @natcomms.nature.com‬ 🧵 1/6

In an exemplary in vitro/in vivo study, 1st authors ‪@btanalikwu.bsky.social‬ & Alice Deshayes showed that dense linear protein arrays (e.g. telomeres) do stop loop-extruding SMCs!
