1/3rd of world has immune memory against TB: definitely. Question is: do these people harbor living Mtb that will reactivate decades later and cause disease? Argument is no, and instead all the disease we see is driven by (relatively) recent exposures to people with active disease

Absolutely agree on challenge and need for TB drugs. But the “1/3 of world has latent TB” isn’t so clear - Behr, Ramakrishnan et al have made some very interesting arguments that it’s a huge misconception:

Also, most incubation steps probably only need to be a few minutes, and you can usually freeze and reuse your antibodies… 🥰

I wish I still had a scan of it but my first western in the Lampson lab was completely black?!? Did I not block? 🤷‍♂️ But I really dug in on Western technique to be able to blot for INCENP, including switching to pH 11 CAPS buffer, and have loved westerns ever since.

Neat trick if you polycolonal ab's suck. Incubate them with fixed cells with a KO of your protein of interest, then spin. Protocol here: www.med.upenn.edu/markslab/ass...

On the left - western blot of B16F10 cells wt and KO for CDK8. Our in house produced antibodies give a lot of unspecific bands. On the right same probes with antibodies preincubated with fixed CDK8 KO cells - there is a specific band and faint unspecific bands, which can be probably eliminated with increase of amount of KO cells.
