✅ Genetic risk for VaD linked to early phenotypic changes, years before diagnosis

✅ Strong age-dependent associations with WMH, depression, blood pressure, glucose, and education

✅ Evidence supports midlife as a key window for prevention

Using data from over 330,000 UK Biobank participants, we:

- Built a polygenic risk score for vascular dementia

- Ran a large-scale, age-stratified PheWAS across 9,000+ traits

- Followed up key associations using two-sample Mendelian randomization

Hi Lorenzo! It's primarily using genetic epi methods - so environmental determinants will be assessed with genetically informed methods like Mendelian randomisation, polygenic risk score analysis, GSEM etc. Hope that helps!
