Very happy to share our new paper now on @medrxivpreprint: “Genetic risk effects on psychiatric disorders act in sets”, a great effort led my PhD student @jolienrietkerk.bsky.social, and performed together with collaborators Andy Dahl, Jonathan Flint, Andrew Schork etc. Thread 1/n

My sincere thanks also to the participants of the datasets without whom the work would not have been possible:
@ukbiobank.bsky.social and #iPSYCH. As well as those who supported me throughout my PhD work, including
@www.helmholtz-munich.de, @pioneercampus.bsky.social MPI Psychiatry and TUM

I am thrilled to see this work coming to fruition after years of hard work and persistence that could not have been possible without the support and mentorship of
@caina89.bsky.social, the help and guidance of Andy Dahl, Andrew Schork, Jonathan Flint and many others.🧵5/n

Applying this framework on five psychiatric disorders, we find evidence for sets of genetic effects within each psychiatric disorders and their comorbidities. Moreover, their shared genetic factors likely capture common confounders rather than disorder-specific effects.🧵4/n

Coordinated Epistasis, as prev. published by
Andy Dahl,Noah Zaitlen pnas.org/doi/10.1073/... tests whether genetic effects on complex traits act in coordinated sets. It is informative of genetic architecture and provides genetic evidence for unrecognized disease subtypes. 🧵3/n

Using information of millions of individuals both through meta-analyses based polygenic risk scores (PRS) and national register-based family genetic risk scores (FGRS) we investigate psychiatric disorders and comorbidities through the Coordinated Epistasis framework. 🧵2/n

‘Genetic Risk Effects on Psychiatric Disorders Act in Sets’, the title of our new preprint on
@medrxivpreprint.bsky.social! This huge collaborative effort advances our understanding of psychiatric genetic architecture and emphasizes the importance of looking beyond additive effects. 🧵1/n; 🧪👩🏽‍🔬🧬
