Together, this study reveals that, despite a shared "cell type", macrophage aging is strongly influenced by its niche (as well as sex-specific inputs). We hope this metaanalysis is useful to others in the field. 4/4

Ella then proceeded to analyze differentially enriched pathways, revealing increased antigen presentation, ECM remodeling, antioxidant responses, and supression of ferroptosis. Finally, she identified 35 genes with consistent regulation across datasets. 3/4

By systematically curating published #transcriptomic datasets of aging mouse #macrophages, Ella tried to answer the question of whether #macrophage aging follows a common blueprint or shows sex- and niche-specific patterns. Intringuingly, niche was the strongest effect on aging trajectories. 2/4

We also performed functional assays to validate our omic findings, and found that indeed, AluJb-overexpressing fibroblasts show remodeled mitochondria metabolism, S-phase accumulation/replication stalling and decreased aggresome load. 3/4

Here, we show that overexpression of AluJb, a transposition-incompetent SINE, in human fibroblasts, leads to profound remodeling of transcriptome, proteome and secretome. Intriguingly, many AluJb-regulated pathways correspond to aging hallmarks (i.e. mitochondria, proteostasis, self-renewal). 2/4
