Structure and mechanism of bactericidal mammalian perforin-2, an ancient agent of innate immunity
Bactericidal perforin-2 assembles an inactivated pre-pore complex and undergoes a 180° reconfiguration for membrane attack.

Thanks for sharing the work. How is it that with the scale of molecules considered (millions) you found only 2 compounds that showed a hint of activity? kRas is a high bar, kudos to your boldness. However - results like this suggest that either the target or the approach should be abandoned.

I did that myself recently. You can use any of the llm based chat tools (Gemini, chatgpt, etc) to help you quickly reformat. Took me only a few iterations with Gemini to hit my mark.

I think the analysts are choosing not to believe what the Lilly folks are predicting. Crazy, I know. When is the last time a big company overstated their future outlook?

That was left as an exercise for the student.

I agree that the right assays are important. However - the larger matter determining success is the speed and quality of the design-make-test cycle. I have seen many early projects suffer because the chemistry could not deliver the molecules in a timely fashion.
