On the theory side, Yuhai Tu, @qiweiyu.bsky.social, and I developed a decomposition of the covariance between expressed RNA molecules into extrinsic and intrinsic components and we introduced a minimal stochastic model for the expression statistics resulting from coupled gene activation.

The experiments utilized a synthetic topologically controllable chromosomal domain. Combined with gyrase and topoisomerase inhibitors, this allowed us to measure how domain formation and DNA supercoiling modulate expression level and correlations between expression of neighboring genes.
