To reroute a cytokine’s signaling, Abhiraman et al. generated soluble adaptors comprising units that bind to two sites on a cytokine linked to units that bind to and compel dimerization/activation [...] bit.ly/4l0nd2G  @stanfordmedicine.bsky.social  @gitaabhiraman.bsky.social

Conversely, we show that cytokine adaptors can convert inflammatory cytokines into immunosuppressive signals. We show conversion of IL-23 or IL-17 into immunosuppressive IL-10 receptor agonists, with potential applications in autoimmune disease.

As proof of concept, we design cytokine adaptors that convert TGF-b or IL-10 into IL-2 receptor agonists to reverse T cell suppression, with potential applications in cancer therapy.

Excited to share this work from the Garcia Lab out today in @naturecomms.bsky.social! We engineer "cytokine adaptors", molecular immune switches that simultaneously block a target cytokine while inducing local activation of alternative cytokine receptors. rdcu.be/ec7GS
Redirecting immune signaling with cytokine adaptors
Nature Communications - Cytokines are immune signaling molecules that are frequently dysregulated in disease. Here, the authors create engineered cytokine ‘adaptors,’ molecular switches...
