Upon reactivation, some memory B cells differentiate into protective antibody-secreting cells. Risley et al. show that the transcription factor T-bet marks a population of memory B cells poised to rapidly differentiate into antibody-secreting cells. They demonstrate that sustained T-bet expression is required for the persistence and function of these effector memory B cells in lymphoid and mucosal sites.
