Ever wondered how 3D chromatin rewires during differentiation? We wanted to understand this process better & distinguish if different processes or elements drive this rewiring. To do so, we used mESC=>NPC differentiation and focused on a single locus. biorxiv.org/content/10.110… 1/17

Fair point. Maybe I'm biased due to my own background & people I've been surrounded by. Also, although I'm not successful by those metrics, so far, I had so many issues during my education/carrer (many due to my background) and I think I'm still here only due to sheer determination.

Really? Somehow I am not surprised by this at all. It is harder to succeed when you come from underprivileged background. Thus, either many change path early on (0 publication) but the ones that persist probably had to overcome lots of struggles = more likely to have higher impact.

This is what I experienced since I started doing science. I always felt I'm better at developing ideas/discussing novelties/troubleshooting when I have a friend/colleague (ideally in the same person) to bounce ideas off. I think we'd all benefit if academia starts encouraging the concept of co-PIs.

We found that only by combining loop extrusion, weak compartmental interactions, and site-specific loop attractions we could successfully simulate ESC->NPC rewiring. These site-specific interactions could be H3K27Ac-driven. However, given that both P and A are 16/17

indicating that while loop-extrusion halting at A seems to contribute to its scaffolding function, the loss of loop-extrusion contribution isn't enough to halt the rewiring.
