I'm pretty sure one of these faces is dead/missing Mrs. Winchester but I don't know which one, so I'm going to say it's this lady. Tragically died by some crazy demon fire. Lady, what are you doing hanging out on the ceiling!

Here we have Sam's long lost son, Daniel. He shows up in the 5th season and tries to get Sam to let him join the family business. Sam grapples with his sudden parenthood but by the time he comes to grips with it at the end of the 5th season, Daniel is tragically killed by the big bad of the season.

Alastair is being leveraged by the big bad, they're holding his family hostage until he completes some contract. Tragically, his family is killed in an episode which causes Alistair to go rogue and betray the big bad.

This isn't just a bad guy this is THE bad guy. The big bad. The one everyone is searching for, but they don't figure out that it's him until late in the series. Maybe he comes along as a minor character for a while spying on our main cast.
