Human SNW1 can rescue loss of fly Bx42. We are now testing variants to determine how they cause microcephaly.

New neurodevelopmental disorder gene SNW1/Bx42. In flies, we show that loss of Bx42 causes reduced brain size and reduced numbers of neural stem cells that divide less.

#SnapshotsinNeuroscience: Larval fruit fly brain
This image from Roy and Link shows neuronal expression of seizure-associated proteins in a third-instar larval fruit fly brain. Image credit: Paula Roy
https://blog.eneuro.org/2025/06/snapshots_larval_drosophila_brain
The image was captured on a Zeiss 710 confocal microscope at 20x using endogenously tagged Sdc-GFP to highlight Sdc (green), ElaV/Alexa Fluor 647 to stain neurons (cyan), and Imp/Rhodamine RedX to show Imp protein (yellow). Image by Paula Roy.

Our group was featured in a local documentary “Fruit flies: decoding rare diseases.” Super excited to see the final product at SLCC on May 2. It was great to work with Aria and the team from SLCC.

We found that Imp binds Syndecan RNA. Expression of Syndecan in an animal with reduced Imp rescued adult seizure phenotypes. So, Imp regulates Syndecan to promote neural development and function.
nikkilink.bsky.social
Since Imp binds RNAs, we wanted to know the neuronal target of Imp that contributes to seizure behavior.
nikkilink.bsky.social
Developmental Imp played a large role in adult seizure behavior since inhibition of neuronal Imp only during developmental stages had nearly the same effect as continuous neuronal knockdown.
nikkilink.bsky.social
We studied the RNA binding protein Imp in neurons and found loss of Imp is associated with increased seizures in adults.
