💙 1 in 8 UK men will develop prostate cancer — but the risk is twice as high for men of African ancestry.

Yet, research in this group is still limited. That needs to change.

[8/8] 📖 If you would like to learn more about why this research is so important for men with advanced #ProstateCancer, you can read more about the project in our latest News & Views article: prostatecanceruk.org/about-us/new...

[7/8] Impact:

🖊️ Tools that analyse tumour transcriptomes can pinpoint which men with advanced #ProstateCancer are likely to benefit from chemotherapy.

💙 This helps men avoid unnecessary chemotherapy and its associated side effects, ultimately enhancing their quality of life.
A man with a happy expression, smiling and enjoying time with his friends.
prostateukprofs.bsky.social
[6/8] Next, the team identified:

✅ In men with advanced #ProstateCancer, PTEN inactivity is associated with shorter survival on hormone therapy but predicts a greater survival benefit from docetaxel compared to PTEN-active tumours.
Graph showing that men with tumours exhibiting PTEN loss are more likely to benefit from combined chemotherapy and hormone therapy treatment.
prostateukprofs.bsky.social
[5/8] First, the study revealed:

✅ The Decipher signature is prognostic and helps identify men with advanced #ProstateCancer who are most likely to benefit from docetaxel.
Graph showing that men with high Decipher scores benefit from combining docetaxel chemotherapy with standard hormone therapy, whereas those with low scores do not.
prostateukprofs.bsky.social
[4/8] 🧬 The team analysed transcriptomes from 1,523 biopsies of men previously enrolled in the STAMPEDE trial using the Decipher signature and a signature that assesses PTEN activity, to evaluate their effectiveness in guiding treatment stratification.
Schematic illustrating the workflow for processing biopsies from men, including central H&E review, tumour macrodissection, microarray hybridization, and quality control analysis.
prostateukprofs.bsky.social
[3/8] 💊 Yet, while docetaxel extends survival for some men, responses vary, and clinicians have limited tools to predict who will benefit.

💡The team explored whether transcriptome analysis tools could identify men likely to benefit from docetaxel, with a particular focus on the Decipher signature.

[2/8] The STAMPEDE trial, led by @prof-nick-james.bsky.social and funded by @cancerresearchuk.org, showed that combining docetaxel chemotherapy with standard hormone therapy significantly improves outcomes for men with advanced #ProstateCancer.
