SiR-XActin, SPY650-Fastact and SiR-actin, along with whole jasplakinolide-like actin ligand based tools are cell permeable small molecules which do not require genetic manipulation. Dissolve in DMSO, add some to the medium and image with your favourite microscope :)

I was impressed with SiR-XActin: similar labeling of F-actin as Lifeact, didn't detectably alter actin dynamics, and because it can remain in solution while imaging, photobleaching is not a major concern (Below, LA required correction, SiR-Xactin did not). Happy to have played a small part in this!

SiR-actin labeling can be cytotoxic and affects actin dynamics - while the new XActin probes do not! Developed by @veselin-nasufovic.bsky.social and perfect for time-resolved and STED imaging without the need for genetic manipulation.

4/5 🔬 We have demonstrated that #𝗫𝗔𝗰𝘁𝗶𝗻 spectral properties can be tuned with other #SiR and #MAP fluorophores covering the important part of the imaging window.

3/5 💡 The key to our success was a careful #cryo-EM structure-driven SAR study of the #XActin ligand, in which we synthesized and tested more than 100 cycic peptides. We minimized cytotoxicity and preserved its binding to F-actin.

Thrilled to share with you our work on covalent probes! By introducing a biocompatible sulfonium-based cleavable linker, we developed a series of fluorescent probes that can label covalently endogenous tubulin in various cell lines.
