Our study presents a systematic characterization of polarized THG (pTHG) signals from individual axons, enabling the derivation of myelin scores in various systems. 🐭🐟
We confirm the sensitivity of THG for myelin in mouse tissue and detect early axon development in zebrafish.

We show that axons exhibit complex pTHG profiles that depend on axon morphology and can't be explained by classical models. We establish a numerical strategy to relate pTHG profiles to axon diameter and myelin thickness, taking into account myelin-induced optical aberrations.
