Thus, epithelial responsiveness is coupled to cell identity, which is governed by the WNT pathway. These findings provide new insights into intestinal epithelial reponsiveness to microbes, which is relevant for our understanding of gut diseases such as IBD.

What causes this switch when intestinal stem cells differentiate? The WNT pathway is a key determinant of intestinal cell identity, but its role in immuity is less well explored. Here we show that activation of the WNT pathway sensitizes differentiated cells to bacterial antigens.

How is this differential responsiveness regulated? Our research indicates that enterocytes do not respond to bacterial antigens as a result of reduced NF-κB pathway activation and also post-transcriptional control of mRNA stability.
