What is the significance of the MAPK signaling pathway in the diagnosis and subtype classification of intervertebral disc degeneration? Yong Liu & coauthors investigated this question: onlinelibrary.wiley....
The bioinformatics analysis workflow of this study is shown in Figure 1. We obtained 20 differentially expressed MAPK-related genes, of which RASGRP4, DUSP3, HSPA1A, HSPA6, IL1R1, KRAS, MAP3K5, RPS6KA1, BRAF, TNF, CACNA1I, and RPS6KA5 were overexpressed in IDD samples, whereas RASGRP1, DUSP14, DUSP2, RRAS2, JUN, PRKACB, RAP1A, and RAP1B were downregulated (Figure 2A,B). We researched the functions and molecular pathways of 20 differentially expressed MAPK-related genes in IDD. Biological process (BP) terms are related to Ras protein signal transduction, positive regulation of MAPK cascades, and regulation of ERK1 and ERK2 cascades; cellular component (CC) terms are related to ficolin-1-rich granule lumen, focal adhesion, and external side of plasma membrane; and molecular function (MF) terms are related to MAP kinase phosphatase activity, GDP binding, and G protein activity (Figure 2C Supplementary Table S2). Nephograms of the stress distribution in the annulus under flexion loading conditions: Comparable stress values can be recorded in both Models 1 and Model 2, and a lower stress value can be observed in Model 3 and Model 4. Therefore, biomechanical significance of articular processes excision and preservation was limited. In contrast, obvious difference in stress values on the cranial motion segment can be observed between models with PLC excision and preservation.

Workflow of this study. A series of analyses were performed to identify candidate bone morphogenetic protein (BMP) genes associated with the pathogenesis of lumbar disc degeneration (LDD). BMP-related genes were extracted from the GeneCards database. Five transcriptome datasets, including LDD patients and healthy controls (HCs), were obtained from the GEO database, and a meta-analysis was conducted to ascertain the differential expression of LDD-associated BMP genes, followed by cell type-specific expression analysis (CSEA). A three-step summary data-based Mendelian randomization (SMR) method was used to integrate the genome-wide association study (GWAS) summaries and cis-eQTL (expression quantitative trait locus)/cis-mQTL (methylation quantitative trait locus) data from blood, and the putative blood BMP genes and their regulatory elements associated with LDD risk were prioritized (p-SMR-multi <0.01; HEIDI p > 0.05). Contrasts a detached nucleus pulposus or contained disk herniation in a degenerated IVD compared to a healthy IVD. In the degenerated disk, there are many possible mechanisms of pain generation. (1) Damaged AF results in proliferation and activation of mechanoreceptors and nociceptors of the sinuvertebral nerve in the AF and nucleus pulposus. (2) Large, herniated disks can lead to mechanical compression and ischemia of surrounding neuronal structures. (3) Proinflammatory biomarkers like macrophages, TNF-a, C-reactive protein, and IL-1 are released as a response to the degenerative IVD and nerve damage.
