In their Opinion article, Samantha Webster @llssam.bsky.social, Remi Brynn and Kate Poole @katepoole.bsky.social consider how perspectives from both cell biologists and biophysicists will be crucial for characterising mechanosensitive ion channels in vivo.

Examples of in vitro measurements (left panel) and the in vivo mechanical landscapes where tissues and cells experience complex compound forces (right panel). In vitro experimental techniques can provide detailed information about specific properties of mechanosensitive ion channels. However, an experimental gap remains in understanding how channels are activated and the consequences of their function in complex in vivo settings.
