🚨 Excited to share our new preprint: Stag2 dependent chromatin remodeling enforces the erythroid-specific Gata1 cistrome. This work was led by Columbia VP&S MD student Varun Sudunagunta (‼️ Program Directors: Varun is applying to IM residencies this year!) www.biorxiv.org/content/10.1...
Stag2 dependent chromatin remodeling enforces the erythroid-specific Gata1 cistrome
The transcription factor GATA1 has pleiotropic hematopoietic functions, particularly in erythroid and megakaryocytic ontogeny. While mechanistic investigations have uncovered many facets of GATA1 biol...

And is it just FLI1? We don’t think so! Actually a HUGE part of this mechanism coming into focus was it helps us unify our prior PU.1 finding and may explain your zebrafish runx1 data that has been the inspiration for so much of our work! journals.biologists.com/dev/article/...
Cohesin-dependent regulation of Runx genes
Runx transcription factors determine cell fate in many lineages. Maintaining balanced levels of Runx proteins is crucial, as deregulated expression leads to cancers and developmental disorders. We con...

Nope! No change in my overall expression by RNA or protein—but a redistribution of the protein towards a much higher chromatin-bound fraction.

And we think this disordering of the FLI1 cistrome is a big part of the mechanism!

👇👇 Check out our first pre-print!
My very first post on 🦋. 1. Do you know that cohesin ring controls how transcription factor (TF) access chromatin? Excited to share my first piece of postdoctoral work where we investigate how Stag2 cohesin contributes to leukemia! 1/10. www.biorxiv.org/content/10.1....
