Wouldn't it be great if we could not only image large connectomic volumes but also completely reconstruct them? And if a whole mouse brain project didn't cost billions?

With the PATHFINDER preprint (www.biorxiv.org/content/10.1...), we preview a future where it doesn't have to.

Cheers to @tobiasackels.bsky.social, with whom we started the correlative workflow, to Anne Bonnin, key in the SXRT datasets that articulate the pipeline, and to Carsten Waltenberg for milling a targeted cylinder containing the neuronal circuits.

Big shoutout to all collaborators. Starting with @yuxinzhang.bsky.social, whose PhD evolved building this story. To @andreas-t-schaefer.bsky.social, core PI in this endeavour, and @apacureanu.bsky.social and Peter Cloetens for the expert insight on X-ray holotomography.

We developed our correlative multimodal imaging pipeline to incorporate Synchrotron X-ray Holographic Nanotomography.
With it we mapped a targeted 0.3mm^3 volumes with subcellular detail, non-destructively.

With sufficient reliability to gather 3 replicates of the same neuronal circuit.

In a nutshell, neurons of the same olfactory circuit in the mammalian brain share firing properties while showing individual traits.

Check out the tweetorial below by @yuxinzhang.bsky.social for the main findings:

My PhD project is out as a preprint!
We combined 2P and synchrotron X-ray to understand mouse olfactory bulb circuits, linking physiology to structure in 3 animals!

#preprint:

We mapped the structure and function of olfactory bulb circuits with in vivo 2-photon microscopy and synchrotron #Xray holographic nanotomography.
