I see this with the VPN at work as well: Cloudflare tends to throw captchas much more frequently. It is what it is.

Speedwise, Proton VPN is fine for that. Just constant checks of my humanity.

Fits perfectly with this not being a study on the effects of drinking coffee but of sudden withdrawal of coffee. The fact that this was published without the actual control group (people who don't drink coffee at all) is quite schocking. Doesn't speak for the journal.

Sorry @erictopol.bsky.social it doesn't. All participants were normal coffee drinkers. Therefore, the coffee-drinker arm is the Control arm. The intervention of going cold turkey increased the hazard of AF by 64% (100/0.61). The whole report is written from the wrong perspective.

Purified proteins? In cultured cells? In an intact organism?

On cells, I faintly recall the mitochondrial outer membrane assay for cultured cells. Just to add more options to this thread 😁

Isn't the biggest issue with BiFC that the probes themselves have a binding affinity that is non-zero?

I would lean towards some form of FRET assay. Ideally using fluorescence lifetime as readout. But I might be a bit biased 😉

Whenever I needed a strongly fluorescent sample that had some structure and depth, I simply grabbed a fresh leaf outside. This was multiphoton, but there should be strong autofluorescence in 1P as well.
