Using long hairpins too for my research right now ☺️ My strat for doing it in Arabidopsis is cloning it with golden gate and putting fluorescent seed as marker. Unique overhangs works best for assembly, doing same overhangs for all fragments (to make inverts) had pretty crap cloning success.

Apparently, bryophytes time and again include microbial genes in their genomic repertoire, and occupy a langer gene space than vascular plants.

Pluripotent, potatoes are a result of root apical meristem doing their thing, and plants can be regrown from the tubers. But the real answer is, depends on the plant.
