And we confirmed this by performing 2-color TIRF imaging of VAMP2-pHluorin and clathrin-mCherry along axons, showing that over 5 minutes, very few exocytic sites were also sites of clathrin pit formation 10/11

So are exocytic events taking place in the same spectrin clearings as clathrin pits formation? Postdoc Christopher helped extending our correlative approach to 2-color SMLM on the @abbelight.bsky.social scope, showing that exocytic sites are distinct from clathrin pit-containing clearings 9/11

Are exocytic events happening in holes of the spectrin scaffold? To answer this question, we developed live-cell / SMLM correlative imaging to link exocytic sites to their nanoscale environment. Yes, exocytosis along the axon shaft occurs in areas devoid of the submembrane spectrin scaffold! 7/11

In our neuronal cultures, we could visualise spontaneous exocytosis of VAMP2-pHluorin containing vesicles not only at presynapses (in magenta below), but also along the axon shaft itself (outside of the pink blobs) 5/11

Seeing exocytosis is harder than endocytosis: faster, no clathrin pits visible in fixed cells, requiring live-cell imaging! We used TIRF to image the release of vesicles containing VAMP2-pHluorin along axons, and pHusion from @stephgupton.bsky.social's lab (doi.org/10.1242/jcs....) to detect it 4/11

What about exocytosis? It would also need access to the plasma membrane - it happens a lot at presynapses that happen to be devoid of the periodic submembrane scaffold… Can exocytosis happen along the axon shaft at sites determined by the actin-spectrin scaffold? 3/11

Awesomeness from the NeuroCyto lab (kudos @wiesner-t.bsky.social for great story)! They found exocytosis in "holes" within the axon's spectrin skeleton, separate from endocytosis sites. Our team mapped structural patterns and disruptions (via #SReD). Thanks for including us @christlet.bsky.social
