I've cut a couple of big moments out of my comic, when I realized that the connective tissue does a better job of moving things forward without it.
It's like with Mr. Show, where a lot of the best jokes come from the in-between sketches. When you take those out you lose the show's specialness.

If starting at the beginning doesn't work, start at the middle or end and work backwards. Write the part you want to get to and then build bridges to and from it.
