Tree no. 15355 Silver birch (Betula pendula) Medium-sized deciduous tree, native to the British Isles. Known for its white bark, which sheds in strips. It has small, light-green, triangular leaves. Male and female catkins are found in the same tree (monoecy), with wind pollination and wind dispersal of seeds. With fast early growth, birch is a useful pioneer species. Birch trees can also improve soil quality by recycling nutrients. Birch is frost resistant and windfirm but planted trees can be vulnerable to birch dieback (caused by fungal pathogens). Naturally regenerated birch – grown from seeds that have fallen from a tree – appear less prone to disease but climatic variables also play a part. Birch woods have a light canopy, encouraging the growth of other plants on the forest floor, and are valuable to wildlife.

(A) Insulin or exercise stimulation initiates a trafficking mechanism termed fusion-guided GLUT4 entry (1, FGG4E), whereby IRVs undergo heterotypic fusion with TfR-positive endosomal recycling compartments (ERC) near the TGN- or ERGIC-like regions. This fusion enables GLUT4 to enter the dynamic recycling endosomal system, where it adopts TfR-like motility and undergoes repeated rounds of trafficking to the PM (2, Traffic). Notably, as long as insulin stimulation persists, GLUT4 remains in this dynamic endosomal loop (3, Recycle) and does not return to its original storage pool, suggesting an active mechanism that prevents retrograde retrieval. (B) Upon cessation of insulin stimulation, GLUT4 is extracted from the recycling pathway through a sortilin- and retromer complex-dependent retrograde trafficking process. This leads to its transient entry into the golgin-97 (also known as GOLGA1)- and golgin-245 (also known as GOLGA4)-positive TGN, where it is transiently retained and subsequently re-packaged into IRVs. Under basal conditions, GLUT4 is statically sequestered via AS160-dependent mechanisms, remaining in a primed state for the next stimulus. Together, both panels illustrate a reversible cycle of GLUT4 trafficking regulated by stimulus-dependent fusion entry and retrograde resetting, balancing dynamic integration with stable retention. Note: this figure emphasizes the TGN-based retrograde trafficking pathway of GLUT4, which is directly supported by our observations involving sortilin, retromer, and golgin-97 and -245, and indicates that the GLUT4 vesicle biogenesis system is enhanced upon differentiation. Nevertheless, we do not exclude the possibility that GLUT4 might also traffic through ERGIC-associated compartments, as suggested by previous studies.

Overview of the LDR. Lysosomal stress and membrane damage are triggered by diverse harmful factors, including physical stressors (e.g. silica and crystals), oxidative stress, lipid peroxidation, amyloid fibrils and bacterial infection. Lysosomal damage leads to the leakage of lysosomal contents such as cathepsins (a type of lysosomal protease) and protons into the cytosol. This causes proteolytic damage, mitochondrial dysfunction, inflammation and potentially cell death, ultimately resulting in accelerated aging and disease (e.g. neurodegeneration, kidney injury). The LDR involves at least three pathways – membrane repair, removal of damaged lysosomes, and lysosome biogenesis and regeneration. The activation of these pathways depends on the extent, duration and severity of the damage (double-headed arrows). Mild or transient stress or damage is rapidly resolved within seconds to minutes through lysosomal membrane repair mechanisms. When the damage exceeds the repair capacity, indicated by persistent rupture or exposure of luminal glycoproteins, the cell initiates selective autophagy of damaged lysosomes (called lysophagy). In addition, when damage is widespread or chronic, and functional lysosomes are depleted, lysosome biogenesis and regeneration occur.
