This has therapeutic implications! We show that low PRPP levels in NUDT5-KO cells prevent the conversion of nucleobase analogs #5FU #6TG into their toxic nucleosidic forms, leading to chemotherapy resistance.

Could the NUDT5 status of a tumor inform treatment?

The NUDT5-PPAT interaction appears to be disrupted by PRPP in a #physiological feedback loop! Indeed PRPP serves not only as the PPAT substrate but also as its main #allosteric activator!

⚙️ High PRPP → dissociation of NUDT5 from PPAT → increased purine synthesis

We show that the interaction of NUDT5 with PPAT prevents PPAT’s full activity and maintains the #PRPP pool, which is the ribose donor common to both purines and pyrimidines!

⚙️ Low NUDT5 → hyperactive PPAT → no more PRPP for pyrimidines

🧩 We made two key observations for the mechanism:
(1) NUDT5 binds to and oligomerizes with #PPAT, the first and rate-limiting enzyme in purine synthesis
(2) PPAT depletion rescues pyrimidine synthesis in NUDT5-KO cells #epistasis

But why pyrimidines?!?

And so, we focused on hit number 4: #NUDT5 (#NUDIX5)!

While we confirmed low pyrimidine levels in NUDT5-KO cells, we were surprized to find an increase in #purines, indicating nucleotide imbalance.

How is this possible, and how is NUDT5 involved?

Nucleotides are essential for life and can be synthesized de novo or obtained through #salvage. Genes involved in de novo synthesis are essential, except when a substrate for salvage is provided.
