-Patient-derived organoids from individuals with Koolen-de Vries syndrome reveal impaired DNA replication and reduced synapse formation, driven by excess R-loops.

Key insights:
-The NSL complex prevents R-loop accumulation at weakly transcribed promoters, safeguarding DNA replication.

-Loss of NSL disrupts S-phase progression and causes replication stress.
