These medications work by blocking the action of the enzyme PDE5, which is responsible for breaking down cyclic GMP (cGMP) in the smooth muscle cells. By inhibiting PDE5, these drugs promote the accumulation of cGMP, leading to vasodilation in the smooth muscle cells of the blood vessels.
