Using scRNA-seq, 3D-hE-gastruloids map directly onto in vivo post-implantation epiblast and early primitive streak.
They express:
🔹 Pluripotency: OCT4, SOX2, PRDM14
🔹 Epithelial/polarity: CDH1, CLDN6/7, PODXL
🔹 PS markers: TBXT, MIXL1, EOMES (but only in a subset)

This model, called “3D‑hE‑gastruloids” is powerfull because it eliminates undefined factors and extraembryonic cells, allowing us to precisely test the signals that govern epithelial identity and PS induction.
Reproducibility is high: >95% of structures form a correct lumen by day 4.

We generated mosaic gastruloids with varying proportion of mutant cells to force differential N-Cadherin expression. Interestingly, the elongation was directly correlated to the proportion of mutant cells, linking gastruloid elongation to cell adhesion. 4/12

Here, we could confirm that gastruloids lacking N-Cadherin self-organize correctly, in fact even better than their wildtype counter part as they form polarized somitic tissue without requiring exogeneous extra cellular matrix such as matrigel. 2/12
