Happy to share the molecular mechanism of activation of kainate-subtype ionotropic glutamate receptor GluK2 in complex with auxiliary subunits Neto2. In both closed/apo and open states, ligand-binding domain (LBD) layer has 2-fold rotational symmetry!

1/x New story - the structure of the Vault cap, at 2.3Å! Matching a beautiful preprint from @sjorsscheres.bsky.social, we show that the cap of the vault particle has C13 symmetry, in contrast to the 39-fold symmetry of the body. Study lead by talented postdoc @huanli00.bsky.social from my group! ⬇️

Interestingly, the vanilloid, melatonin, and mucolipin subfamilies of TRP channels follow a similar pattern of conformational changes upon opening, but inactivation occurs differently across subfamilies. (5/5)

To date, several structures of TRP channels in complexes with interacting proteins were published: TRPV5/6 and TRPC4 with calmodulin, TRPV4 with RhoA, TRPM3 with Gβγ, and TRPC5 with Gαi3. (4/5)

In the apo state, in high-resolution structures, many of these ligand-binding sites are occupied by annular lipids. However, not all lipid sites were reported to be occupied by ligands, so these new sites might be interesting to explore pharmacologically. (3/5)

We compared all structures of TRP channels released on the RCSB by 2024-08-14 and identified 40 distinct ligand-binding sites. (2/5)
