3/3 This advice brought to you by our conclusion that no antibody will specifically detect Smoothened in embryo lysates. We screened every ab available. We have resorted to IP/westerns w/IgG control (essential). The blots are still dirty, but at least we can be confident about signal specificity 😵💫😥

2/3 Crucial: Always have a good positive and negative control - like a lysate from a knockout embryo or cell line (negative) and a cell line expressing exogenous protein (positive) 🤓 Do your experimental bands line up with controls? If not, do not trust the band.
