🎉 New paper out in Science!
In collaboration with Marc Therrien’s lab, we show how oncogenic BRAF mutants adopt a pre-active conformation, escaping auto-inhibition and driving RAS–ERK pathway activation in cancer.

BRAF oncogenic mutants evade autoinhibition through a common mechanism
Uncontrolled activation of the rat sarcoma (RAS)–extracellular signal–regulated kinase (ERK) pathway drives tumor growth, often because of oncogenic BRAF mutations. BRAF regulation, involving monomeri...

This one is a bit of a departure from the usual and definitely a work in progress!

We found that by using ab initio reconstruction at very high res, in very small steps, we could crack some small structures that had eluded us - e.g. 39kDa iPKAc (EMPIAR-10252), below.

New research in Science greatly expands the potential target scope of molecular glues and should stimulate the development of new small molecules that can selectively target therapeutically relevant proteins for degradation.

This illustration shows the interface of the kinase NEK7 (light blue) and the E3 ubiquitin ligase substrate receptor cereblon (teal). The interaction is facilitated by the small molecule MRT-3486 (orange).
