The findings contribute to the growing body of evidence that manipulation of host pathways, in particular cellular cholesterol homeostasis & #mTOR signalling axis ➡️ a viable strategy to restrict HAdV (human adenovirus) infection.
Mode of antiviral action of itraconazole and Ku-63794 in HAdV-infected cells. (1) After binding of HAdV to its entry receptor, (2) receptor-mediated endocytosis is induced, leading to (3) endosomal escape and (4) virus uncoating. The virus is shuttled to the nucleus, where (5) transcription of early virus genes is initiated, followed by (6) viral protein synthesis and (7) viral replication. (8, 9) The switch from early to late viral protein synthesis triggers the (10) assembly of new virions and (11) virus release. During infection, HAdV hijacks and activates the host cell machinery, including the activation of the PI3K/Akt/mammalian target of rapamycin (mTOR) pathway. The mTOR signalling is activated through two viral proteins, E4 ORF1 and E4-ORF4, which activate mTOR signalling directly (via PI3K) or indirectly (via PP2A), thus promoting cell growth, cellular and viral protein synthesis and potentially suppressing cellular antiviral response. Itraconazole interferes at two different stages of HAdV infection. The early antiviral effect of itraconazole is mediated by cholesterol sequestration in endolysosomes that hinders virus uncoating and thereby promotes endolysosomal degradation of HAdV. Additionally, itraconazole inhibits the virus-promoted activation of mTOR through activation of AMPK. AMPK inhibits mTOR signalling, thus affecting viral protein synthesis and survival of HAdV-infected cells (induces autophagy of infected cells to clear virus infection). Ku-63794 interferes with mTOR activation by directly inhibiting mTOR and by inhibiting Akt, a protein upstream of mTOR. Akt inhibition promotes TSC1/2 activation, resulting in Rheb inhibition and inactivation of the mTOR pathway. Both mechanisms contribute to lower virus particle production and decreased infection rates.

The rare clot condition, Vaccine-Induced Immune Thrombotic Thrombocytopenia (VITT), has primarily been associated with 1st dose of the adenovirus-based AstraZeneca & Janssen 💉

PNAS RESEARCH ARTICLE MICROBIOLOGY As SARS-CoV-2 variants of concern (VoCs) that evade immunity continue to emerge, next-generation adaptable COVID-19 vaccines which protect the respiratory tract and provide broader, more effective, and durable protection are urgently needed. Here, we have developed one such approach, a highly efficacious, intranasally delivered, trivalent measles-mumps-SARS-CoV-2 spike (S) protein (MMS) vaccine candidate that induces robust systemic and mucosal immunity with broad protection. This vaccine candidate is based on three components of the MMR vaccine, a measles virus Edmonston and the two mumps virus strains [Jeryl Lynn 1 (JL1) and JL2] that are known to provide safe, effec- tive, and long-lasting protective immunity. The six proline-stabilized prefusion S protein (preS-6P) genes for ancestral SARS-CoV-2 WA1 and two important SARS-CoV-2 VoCs (Delta and Omicron BA.1) were each inserted into one of these three viruses which were then combined into a trivalent "MMS" candidate vaccine. SARS-CoV-2 | intranasal trivalent vaccine | MMR vaccine The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has resulted in tremendous morbidity and mor- tality worldwide and continues to threaten all aspects of our society (1, 2). As of May 2, 2023, more than 6.9 million deaths and 764 million infected cases have been documented. Several types of SARS-CoV-2 vaccines have been approved for use in human including messenger RNA (mRNA), adenovirus-vectored, and subunit vaccines (3). Most of them use the prefusion spike (S) protein stabilized by two prolines (preS-2P) as the immunogen (4-6). Following two intramuscular injections, these vaccines are successful in preventing severe disease and death (4-6). The continuous emergence of new SARS-CoV-2 variants of concern (VoCs) has made this pandemic difficult to halt. Since the pandemic began, several VoCs including the Alpha (B.1.1.7), Beta (B.1.351), Gamma (P.1), Delta (B.1.617.2), and, more recently, the dominant Omicron (B.1.1.529/BA. 1) and its subvariants (BA.2, BA.2.12.1, BA.4, BA.5, and XBB.1.5) have emerged (7, 8). The Omicron BA.1 variant was first detected in South Africa in November 2021 and spread globally (9). Compared to earlier variants, the Omicron BA.1 S protein harbors the highest number of mutations in the receptor-binding domain (RBD), markedly reducing its sensitivity to neutralizing anti- bodies (NAbs) induced by the current COVID-19 vaccines (10).
