This can have major consequences for drug design: the internal unbinding pathway points toward the cell membrane, so this path may increase the chances of protein–ligand rebinding due to the presence of the membrane as a barrier for free diffusion of the ligand. (3/3)

We characterized for the first time paths for agonist and antagonist dissociation from the Toll-like receptor 8.
Two paths were found.
Interestingly, some ligands have clear path preferences: the agonist uses only the external path, and the cationic antagonists use only the internal path. (2/3)
