Absolutely: checkpoints, self-recognition, and now Tregs all getting up but the discovery of the cellular mediators (and where they come from) continually snubbed. Was told to me that the Robert Good tolerance fiasco may have tainted the field as he would likely have been included in a thymus gong.

A small primer on the #NobelPrize awarded to Mary E. Brunkow, Fred Ramsdell and Shimon Sakaguchi today. This prize was for combining two separate fields of immunology research - genetic research on IPEX and immunology research of regulatory T cells (#Tregs), with enormous impact on biology/medicine

Moreover, despite the increase in IFNg, we also found that thymic epithelial cells (TECs) actually decreased their MHCI expression after damage, making them vulnerable to NK-mediated killing. Given the central importance of TECs for all thymic function, their killing after damage inhibits repair.

But to our surprise, although upregulated in response to IL-18, this inhibitory effect was not mediated by IFNg. Instead, both perforin and granzyme could limit thymic regeneration, but only perforin was regulated by IL-18.

Using a slew of mouse strains and assays including imaging, single-cell sequencing, parabiosis, and more, David found that IL-18 released by hematopoietic (DC/macrophage) and non-hematopoietic (mesothelial) cells activated NK cells (but not ILC1) limiting repair via their cytotoxic function.

However, while mice deficient for IL-1 signaling showed no phenotype, those deficient for IL-18 exhibited significantly improved recovery, suggesting that IL-18 plays a suppressive role in thymic repair. Once we had this finding, David was off to the races to uncover all the mechanistic details.
