In early fetal development, blood cells are made in the liver & spleen, but by the time of your birth, almost all your blood cells were being made by hematopoietic stem cells (HSCs) in the medulla of your wet bones.

In adults, specific bones do most of the "heavy lift" in blood cell production.

5/ There were also similar cells found growing in a new 3D culture system.🤔 A new cell model?💡It activated many marker genes! We would’ve never considered this model because it was designed to expand HSCs from PBMCs, completely unrelated to ILD macrophages! 🙌

• Inducible deletion of aCDase in HSCs promotes MMP-1-mediated collagen degradation and ECM remodeling, leading to fibrosis regression.
• Developed a novel small-molecule aCDase inhibitor, compound 5A, demonstrated efficacy in enhancing ECM degradation and resolving fibrosis in two mouse models.

Exploiting elevated mitochondrial membrane potential, we used long-chain alkyl-TPP molecules (ex. MitoQ) that selectively accumulate in the mitochondria. These caused reduced mitochondrial respiration, apoptosis, and ablated the competitive advantage of Dnmt3a-mutant HSCs.

Led by Gunnar Thompson. We're developing a model of the hypoxic bone marrow that supports hematopoiesis. We show hypoxia & angiocrine signals produced by the marrow vasculature shape HSCs. Strikingly, we must mitigate oxygen shock during isolation using cyclosporin A (a first for in vitro culture)!
