Do other PARPs label substrates for degradation as well? In which conditions is this most relevant? How do ADP-ribose and ubiquitin connect to regulate degradation?

This is a very exciting finding, as it connect well with several recent publications that indicate that proteins modified by PARP7 are degraded. For example: doi.org/10.1016/j.ce...

Q for the hive-mind 🧠: What transfection reagents do people like for big constructs these days? (I am thinking #HSV encoding BACs, ~170 kb). We've recently had poor performance with Trans-IT LT1 and Lipofectamine 2000, our two go-tos. Target cells are U2OS. Suggestions or RTs greatly appreciated 🙏
