Excited to share our latest preprint "Capsid Restructuring Activates Semi-Conservative dsRNA Transcription in Cystovirus ɸ6". We show that nucleotide-triggered disassembly of the outer capsid layer is followed by expansion of the inner layer and the stepwise assembly of transcription complexes.

Session 9 Aude Bernheim Unveiling highly diverse anti-phage systems. Evolution of immunity from bacteria to human to discover new immune actors. Why would bacterial anti phage natural agents be useful for antiviral therapeutics? Only 3% biosynthetic gene clusters known. #LorneProteins2025
Lanthipeptide BGCs are found in defence islands. Do lanthiphages confer anti-phage defence? Engineered lanthiphage expression clones directly target the virus; restricted to actinobacterial defence systems. Can they be active against human enzymes?

Ancestral immunity from bacteria to predict novel immune mechanisms in human? But - long evolutionary distances. Evolution is key to understanding what's going on. Iterative/structural searches uncover many false positives. Gof/Lof/already known. SIR antiphage proteins mediate immunity via the TLR pathway. SIR2 domains in antiphage systems. Also present in euk immunity?

SIR2 across domains of life - non-immune sirtuins in one part of the tree, SIRim domain containing part of the tree. Innate immunity? SIRanc. Hypothetical role based on evolution. KO SIRanc in mice no inflammatory response to bacterial infection. Cells less resistant to viral infection.
