Phosphorylation on tyrosines control key pathways in immunity, cancer, and metabolism. For the first time, we can now design proteins that specifically recognize individual phosphotyrosines, even in disordered regions. (1/8)

When I started working on ion channels 20+ yrs ago, I could have never imagined how many channels there are to explore! Now I’m delighted to be heading to thematic meetings on two of our favorite channels in the span of just weeks - can’t wait to catch up with the ASIC and NALCN communities!

Thanks so much for contributing to this!!

A huge thank you to everyone involved and of course @lundbeckfonden.bsky.social, @carlsbergfondet.dk and DFF for funding.

Collaborator @amelierocks.bsky.social complemented this with predictions of protein stability and conservation scores. In combination with functional work this establishes a framework for classifying suspected disease variants, which we hope will aid assessment of NALCN variant pathogenicity

@chowhanchua.bsky.social implemented use of a combination of multiple parameters derived from two-electrode voltage-clamp electrophysiology recordings. These allow us to classify LoF and GoF phenotypes and to provide mechanistic insight into the molecular mechanism underlying specific variants.
plesslab.bsky.social
De novo and inherited NALCN variants can lead to severe developmental syndromes. Although variants of uncertain significance or presumed pathogenic variants have been studied in heterologous systems before, there is no generally accepted framework on how to assess or predict variant pathogenicity.

After years in making, I am glad to be able to share our attempt on establishing a framework for combined functional and computational assessment of variant pathogenicity in the sodium leak channel NALCN:

Deadline Aug. 15th 2025

villumfonden.dk/en/group/cha...
Villum Foundation wishes to encourage the exchange of scientific ideas between the US and Denmark. Villum Sabbatical supports research stays of up to one year at a Danish university. Researchers at the associate professor or professor level based at a US university may receive up to 1 MDKK to cover their salary (in part or in full), salary of accompanying group members (postdocs and PhD students), travel and research costs associated with the stay. Applications are only accepted from the head of the department at the Danish host institution.

If a visiting researcher subsequently decides to relocate to Denmark, they are encouraged to apply for one of Villum Foundations’s investigator grants (Young Investigator, Ascending Investigator, Investigator)

“It’s the company that matters!”

This is certainly true for acid-sensing ion channel 1a, whose function is modulated by the presence of monocarboxylate transporter 1

All was made possible by the drive & curiosity of the fantastic @mette-h-poulsen.bsky.social
