Inflammasome signaling in astrocytes modulates hippocampal plasticity
The function of immune pathways in homeostatic neurobiology remains underexplored.
Zengeler et al. find that the inflammasome complex, typically associated with immune
responses, is involved in memory...

Direct microglia replacement reveals pathologic and therapeutic contributions of brain macrophages to a monogenic neurological disease
Hematopoietic stem cell transplant is the gold standard for treatment of globoid cell leukodystrophy, also called Krabbe disease. Aisenberg et al. define the molecular signature of disease-associated macrophages (globoid cells) and demonstrate that brain-specific high-efficiency macrophage replacement in the twitcher model of Krabbe disease improves neuropathology, macrophage dysfunction, and survival.

Monocytes can efficiently replace all brain macrophages and fetal liver monocytes can generate bona fide SALL1+ microglia
Replacing brain macrophages holds substantial therapeutic promise but remains challenging. Bastos et al. uncover the requirements for monocytes to replace microglia and border-associated macrophages. They show that monocyte ontogeny shapes brain macrophage identity, with fetal monocytes acquiring a true microglial state. Using xenotransplantation and Alzheimer’s patient data, they identify human monocyte-derived microglia. These findings are important for microglia replacement therapies.

Spatiotemporal Analysis of Remyelination Reveals a Concerted Interferon-Responsive Glial State That Coordinates Immune Infiltration
Remyelination, the process by which axons are re-encased in myelin after injury, is a critical step in restoring brain function, yet the dynamics from initial injury to repair remain poorly characteri...
