Transcriptome profiling of peripheral blood mononuclear cells from highly susceptible adult cattle infected with a virulent strain of Babesia bovis
CONCLUSIONS: In this RNA-Seq analysis, we identified genes that are up- and downregulated in response to acute B. bovis infection. Gene expression of IL-10, along with that of the inflammatory cytokines IL-1β, TNFα and IL-18, suggests a non-protective response to B. bovis at 10 dpi. These results enhance our understanding of the molecular interactions between Babesia and the host immune system.

A convenient single-cell assay for the newly synthesized transcriptome reveals FLI1 regulon downregulation during T cell activation
Lyu et al. present a convenient and accessible method for profiling of both regular and newly synthesized transcriptomes in single cells. Using this approach, they characterize the temporal dynamics of gene expression during T cell activation and identify FLI1 as a master transcription factor downregulated upon T cell stimulation.

Here, we show that overexpression of AluJb, a transposition-incompetent SINE, in human fibroblasts, leads to profound remodeling of transcriptome, proteome and secretome. Intriguingly, many AluJb-regulated pathways correspond to aging hallmarks (i.e. mitochondria, proteostasis, self-renewal). 2/4

Although synaptic mRNAs represent only a fraction of the templates for the synaptic proteome - with many templates residing in the soma or in non-synaptic regions of dendrites and axons - the synaptic transcriptome reflects the synapse diversity captured by the proteome equally well.

Integrating transcriptome and proteome data, we identified ~90 genes with significantly correlated RNA-protein ratios across regions, involved in synaptic vesicle dynamics, receptor signaling, & calcium regulation, suggesting a key role for local protein synthesis in maintaining protein copy number.

Chemical synapses across the brain are highly diverse and it is well established that this manifests at the molecular level. Previous work has primarily focused on the proteome, but here we extend this view and describe another layer of synapse diversity – synaptic transcriptome diversity.

The quantitative genetics approach allowed us to predict evolutionary changes in gene expression at the whole transcriptome level.

With experimental evolution of replicated lines in a stressful hot and dry environment, we could verify that gene expression evolved as predicted after 20 generations.
