📢A new effector-triggered immunity role for GBP1! No place for pathogens to hide now that GBP1 can detect extracellular pathogens that hijack the actin cytoskeleton. Surprise*suprise* GBP1 does this independently of LPS! 🧵

So excited that this study is now online! It's been a pleasure to work with BioAge Labs and our collaborators on this story. We characterise a new class of #NLRP3 inhibitors with translational potential for so many inflammatory diseases.
jem.org
#NLRP3 is a target for antiinflammatory therapies & can be inhibited by the tool compound MCC950. Wilhelmsen, @colllab.bsky.social et al characterize new small-molecule inhibitors, BAL-0028 & BAL-0598, that have a distinct mechanism of action & binding site. rupress.org/jem/article/...

#InterferonPower! Thrilled for our latest work @cp-cell.bsky.social! With @danielboehmer.bsky.social, we dug into tons of papers & created what we hope will be a go-to resource for immunologists & non-immunologist about type I, II, III (& IV😉) #interferons! Stimulating immune cells in vitro is a common experimental lab model. We profiled 150K blood immune cells treated with 11 different stimuli to compare the effects. The data are freely available for researchers. See the preprint for our findings and to access the data 🧪 doi.org/10.1101/2025...

Fantastic work by the Coers lab! GVIN1 (finally) joins the club of antimicrobial GTPases.

Exciting work from neighbor Jörn Coers at Duke identifying a new class of host defense GTPases. Out of a genetic screen they identify the human Giant GTPase GVIN1 (previously annotated as a human pseudogene) in coatomer-based restriction of Burkholderia motility. 1808 aas long!
Human giant GTPase GVIN1 forms an antimicrobial coatomer around the intracellular bacterial pathogen Burkholderia thailandensis
Several human pathogens exploit the kinetic forces generated by polymerizing actin to power their intracellular motility. Human cell-autonomous immune responses activated by the cytokine interferon-ga...

We report today that the structurally complex PAMP LPS can be broken down to a simple pattern that is detected by the innate immune receptor caspase-11. This pattern is a lipid containing a negative charged headgroup + acyl chain ≥14 carbons.

LPS binding caspase activation and recruitment domains (CARDs) are bipartite lipid binding modules
A bacterial sensory protein detects self- and nonself-lipids by similar mechanisms.

Have you ever wondered whether pyroptotic corpses have functions beyond the grave? We made the surprising discovery that pyroptotic corpses are flagged by #filopodia to alert #DendriticCells they need autopsy. A short Bluetorial (1/13)👇with movies!

Pyroptotic cell corpses are crowned with F-actin-rich filopodia that engage CLEC9A signaling in incoming dendritic cells
